目的:探寻与膀胱癌相关的长链非编码RNA,并明确它们在膀胱癌细胞系中的表达状况。方法:用lncRNA双通道表达谱芯片分析4对尿路上皮癌组织及癌旁正常尿路上皮的lncRNAs表达差异,筛选出差异表达的候选lncRNAs,并在膀胱癌细胞系(T24,RT4,BIU-87,EJ)、正常细胞系和其他肿瘤细胞系中,利用实时荧光定量PCR反应(Real-time PCR)进行验证。结果:根据芯片结果,挑选候选lncRNAs共6条,包括DUXAP9、DUXAP10、LINC01296、UCA1、PVT1、MEG3,进行细胞系表达验证。DUXAP9、DUXAP10和LINC01296在膀胱癌细胞系中显著特异性高表达。UCA1在胰腺癌、膀胱癌和肝癌各细胞系中均高表达,PVT1在膀胱癌细胞系中未见特异性高表达,MEG3在膀胱癌细胞系表达亦不高。结论:我们首次报道了DUXAP9、DUXAP10和LINC01296在膀胱癌细胞系中特异性高表达状况,与已报道的UCA1、MEG3、PVT1相比,这三个lncRNAs可能具有更好的膀胱癌特异性。
Objective: To explore long-chain non-coding RNAs associated with bladder cancer and clarify their expression in bladder cancer cell lines. Methods: The expression of lncRNAs in 4 pairs of urothelial carcinoma tissues and adjacent normal urothelium was analyzed by lncRNA two-channel expression profiling. The differentially expressed candidate lncRNAs were screened and expressed in bladder cancer cell lines (T24, RT4, BIU- 87, EJ), normal cell lines and other tumor cell lines using real-time PCR. Results: Six candidate lncRNAs were selected based on the chip results, including DUXAP9, DUXAP10, LINC01296, UCA1, PVT1 and MEG3 for cell line expression verification. DUXAP9, DUXAP10 and LINC01296 were significantly and specifically expressed in bladder cancer cell lines. UCA1 is highly expressed in all cell lines of pancreatic cancer, bladder cancer and liver cancer, PVT1 is not highly expressed in bladder cancer cell lines, and MEG3 is not expressed in bladder cancer cell lines. Conclusions: We first reported the specific high expression of DUXAP9, DUXAP10 and LINC01296 in bladder cancer cell lines. These three lncRNAs may have better bladder cancer specificity than the reported UCA1, MEG3 and PVT1.
