目的:研究四分子交联体5(TM4SF9)在大肠侧向发育性肿瘤(laterallyspreadingturmor,LST)、SW480和Lovo细胞中的差异表达,以及TM4SF9表达差异与三个细胞株黏附能力的关系.方法:用基因芯片技术研究3个细胞株的共同差异表达基因,从中筛选出TM4SF9作为研究对象,用半定量RTPCR技术验证基因芯片检查结果,用黏附实验研究3个细胞株黏附性的差异.结果:基因芯片检查发现TM4SF9在3个细胞株中都有表达,Lovo细胞株表达最强,LST表达最弱,半定量RTPCR检查与基因芯片检查结果相符,黏附实验显示TM4SF9表达水平最高的细胞黏附性最强.结论:TM4SF9在LST,SW480,LOVO3个细胞株中存在差异表达,黏附性较强的细胞株表达较高,其与LST特性的关系值得进一步研究.
Objective: To investigate the differential expression of TM4SF9 in laterally metastasized tumor (LST), SW480 and Lovo cells and the relationship between the expression of TM4SF9 and the adhesion ability of three cell lines.Methods: Gene chips were used to study the common differentially expressed genes of the three cell lines, TM4SF9 was screened out as the research object, the gene chip test results were verified by semi-quantitative RTPCR technology, and the adhesion experiments were used to study the adhesion differences of the three cell lines.Results: Chips showed that TM4SF9 was expressed in all three cell lines, Lovo cell line was the strongest, LST was the weakest, and semi-quantitative RTPCR was consistent with that of microarray. Adhesion showed the strongest cell adhesion of TM4SF9 .Conclusion: TM4SF9 is differentially expressed in 3 cell lines of LST, SW480 and LOVO, the cell lines with strong adhesion are highly expressed, and its relationship with LST properties deserves further study.
