Ras蛋白是调节细胞生长、增殖和分化的重要元件。当Ras变异时,可引起细胞的异常增殖,导致肿瘤的发生。胞外信号调控激酶(ERK)是Ras下游效应子,在恶性肿瘤细胞的生长和预后中扮演重要角色。近来研究证明,在Ras和ERK之间还存在其他相关的效应子,相互之间构成了一条完整的Ras/Raf/MEK/ERK信号通路,在食管肿瘤的发病过程中起着重要作用,部分针对该通路的抗肿瘤药物已在临床中应用。对于Ras/Raf/MEK/ERK信号通路中分子机制的进一步理解,可为制定更有效的食管肿瘤靶向治疗方法创造条件。本文就该信号通路在食管癌中的作用及药物靶点做一综述。
Ras protein is an important element that regulates cell growth, proliferation and differentiation. When Ras mutation, can cause abnormal proliferation of cells, leading to the tumor. Extracellular signal-regulated kinase (ERK) is a Ras downstream effector that plays an important role in the growth and prognosis of malignant cells. Recently, studies have shown that there are other related effectors between Ras and ERK, forming a complete Ras / Raf / MEK / ERK signaling pathway between them and play an important role in the pathogenesis of esophageal neoplasms. The pathway of antineoplastic drugs has been clinically used. Further understanding of the molecular mechanisms of the Ras / Raf / MEK / ERK signaling pathway may provide the basis for more effective esophageal cancer targeted therapies. This article reviews the role of this signaling pathway in esophageal cancer and its drug targets.
