小檗碱对2肾1夹致心肌肥厚大鼠心室重构的影响

目的观察小檗碱对2肾1夹模型大鼠心肌肥厚的抑制作用及对细胞外基质胶原含量的影响。方法采用2肾1夹法制备大鼠心肌肥厚模型,造模4周后根据动物收缩压水平将手术动物均匀分为:模型组,小檗碱5、10、20 mg/kg组,卡托普利45 mg/kg组,另设假手术对照组。连续灌胃给药6周。实验结束时,测定心重指数、左心室肌组织胶原含量和血管紧张素Ⅱ含量。结果小檗碱连续给药6周使2肾1夹模型大鼠心重和心重指数降低,左心室组织胶原含量显著下降,血管紧张素Ⅱ含量也有所减少,作用呈一定的剂量依赖关系。结论小檗碱可能通过抑制心肌间质纤维化而改善心室重构,其作用与对AngII等神经内分泌因素的调节有关。 Objective To observe the inhibitory effect of berberine on myocardial hypertrophy in rats with 2 kidneys and 1 clip and the effect on collagen content in extracellular matrix. Methods The rat model of cardiac hypertrophy was established by two-kidney and one-clip method. After 4 weeks of modeling, the surgical animals were evenly divided into the model group and berberine groups 5, 10 and 20 mg/kg according to the systolic blood pressure of the animals. Lee 45 mg/kg group, another sham control group. Continuous gavage for 6 weeks. At the end of the experiment, the heart index, collagen content in the left ventricular muscle and angiotensin II content were measured. Results The continuous administration of berberine for 6 weeks reduced the heart and heart index of the 2-kidney 1 rat model, reduced the content of collagen in the left ventricular tissue, and decreased the content of angiotensin II. The effect showed a dose-dependent relationship. Conclusion Berberine may improve ventricular remodeling by inhibiting myocardial interstitial fibrosis, which is related to the regulation of neuroendocrine factors such as AngII.
