目的通过测定生长期内不同月份纹党皂苷的含量变化,研究纹党品质形成的变化规律。方法采用人参皂苷含量测定法,用甲醇超声提取后以正丁醇萃取,萃取液回收正丁醇后,残渣用甲醇定容;精密取定溶液低温挥干,加1%香草醛-冰醋酸0.5ml,加入高氯酸0.5ml,60℃下静置反应30min,冷水浴2min后,加入5ml冰醋酸,在540nm处测定总皂苷含量。结果纹党生长期内不同月份总皂苷含量4月含量最高,8月增长幅度最大,9月开始下降。结论纹党总皂苷含量受温度和季节的变化影响较大,纹党皂苷含量积累符合植物开花期、结果期成长的变化规律。
OBJECTIVE To study the variation of quality of Lycium barbarum L. by determining the content of Lycopodium saponin in different months of growth period. Methods The ginsenoside content was determined by ultrasonic extraction with methanol and extracted with n-butanol. After the n-butanol was recovered from the extract, the residue was denatured with methanol. The precision of the solution was evaporated to dryness under low temperature. The solution was added with 1% vanillin-acetic acid 0.5 ml, adding perchloric acid 0.5ml, standing reaction at 60 ° C for 30min, cooling water bath for 2min, adding 5ml glacial acetic acid, measuring the total saponin content at 540nm. Results During the growing season, the content of total saponin in different months was the highest in April, the largest increase in August and the decrease in September. Conclusions The content of total saponin of Lycoris radiata is greatly affected by the changes of temperature and season. The content of Lycium saponin in Lycoris radiata conforms to the regular pattern of the growth of plants during flowering and fruiting phases.
