以芦丁为对照品,选择510 nm为测定波长,580 nm为参比波长,采用双波长分光光度法测定洋葱中黄酮含量。试验结果表明,芦丁在0.008~0.040 mg/mL质量浓度范围内,与吸光度差值ΔA呈良好的线性关系,回归方程为ΔA=5.95C+0.007 3,相关系数r=0.998 9,检出限为0.075 mg/L,测定方法的平均回收率为99.79%,RSD=1.30%。测得洋葱中黄酮含量为1.58 mg/g。试验建立的方法操作简便,稳定可靠,为洋葱的开发和检测提供了依据。
Using rutin as the reference substance, the wavelength of 510 nm was selected as the reference wavelength and the wavelength of 580 nm was used as the reference wavelength. The content of flavonoids in onion was determined by dual-wavelength spectrophotometry. The results showed that rutin had good linearity with the difference of absorbance ΔA in the range of 0.008 ~ 0.040 mg / mL, the regression equation was ΔA = 5.95C + 0.007 3, the correlation coefficient was 0.998 9, the detection limit 0.075 mg / L, the average recovery of the assay was 99.79%, RSD = 1.30%. Flavonoids content of onion was measured at 1.58 mg / g. The method established by the experiment is simple, stable and reliable, and provides the basis for the development and testing of onion.
