目的:探讨己酮可可碱(PTX)是否通过对血清肿瘤坏死因子α(TNF-α)的调节而发挥对大鼠胶原诱导性关节炎(CIA)的保护作用。方法:建立大鼠CIA模型后随机分为正常对照组、生理盐水CIA模型组、PTX治疗组。分别观察各组大鼠的体重、后足容积,并对关节炎的肿胀程度进行评分;用ELISA检测不同时间点各组血清的TNF-α水平。结果:与CIA组相比,PTX治疗组大鼠的炎症得到了较好的控制。第48天时PTX组的TNF-α水平与正常对照组的TNF-α水平没有统计学差异(P>0.05),但与CIA组相比显著降低(P<0.05)。结论:PTX对大鼠CIA关节炎具有缓解症状、抑制炎症肿胀的作用,PTX能够显著降低血清TNF-α水平。PTX可能是通过抑制血清TNF-α水平来发挥抗炎作用。
OBJECTIVE: To investigate whether pentoxifylline (PTX) protects against collagen-induced arthritis (CIA) in rats by modulating serum tumor necrosis factor-α (TNF-α). Methods: The rat CIA model was established and randomly divided into normal control group, saline CIA model group and PTX treatment group. The body weight, the volume of the hindfoot in each group were observed, and the degree of swelling of the arthritis was scored. The levels of TNF-α in each group were detected by ELISA at different time points. Results: Compared with CIA group, inflammation in PTX treatment group was well controlled. The level of TNF-α in PTX group was not significantly different from that in normal control group on day 48 (P> 0.05), but significantly lower than that in CIA group (P <0.05). CONCLUSION: PTX can relieve symptoms and inhibit the swelling of inflammation in rat CIA arthritis. PTX can significantly reduce the level of serum TNF-α. PTX may exert anti-inflammatory effect by inhibiting serum TNF-α level.
