选取健康的性成熟雄性宽体沙鳅,运用JSM 6510LV型扫描电镜、H-7500型透射电镜及Motic-BA210数码显微镜分别观察了宽体沙鳅精子的超微结构及不同浓度Na~+、K~+、Ca~(2+)对其精子活力的影响。结果显示,宽体沙鳅精子头部圆球形,无顶体,细胞核后端有一植入窝凹陷,凹陷深度为细胞核长径的1/6。中片由中心粒复合体和袖套组成。中心粒复合体分为近端中心粒和基体,两者呈“L”型排列;袖套呈两侧不对称分布,一侧狭长,另一侧肥厚。尾部主要由轴丝组成,为典型的“9+2”型双联微管结构,微管动力蛋白臂明显。以Na Cl、KCl和Ca Cl_2浓度分别为75 mmol·L~(-1)、0.5 mmol·L~(-1)和5 mmol·L~(-1)作为宽体沙鳅精子的激活介质,效果最佳。建议实际生产中选取合适的激活介质进行人工授精。
The healthy male mature Hirudo broadleaf carotis, JSM 6510LV scanning electron microscopy, H-7500 transmission electron microscopy and Motic-BA210 digital microscope were used to observe the ultrastructure of S. japonicum spermatozoa and different concentrations of Na ~ +, K ~ +, Ca ~ (2+) on sperm motility. The results showed that the head of Sphaerothera mairei had a spherical, acrosomal-shaped head with an implanted nest recess at the posterior end of the nucleus. The depth of the recess was 1/6 of the long axis of the nucleus. The tablet consists of a centriole complex and cuff. The centriole complex is divided into proximal centriole and matrix, both of which are arranged in “L” shape. The sleeve is asymmetrical on both sides, one side is long and the other is hypertrophic. The tail is mainly composed of axial wire, which is a typical “9 + 2 ” double-type microtubule structure with obvious microtubule motility protein arm. The concentrations of NaCl, KCl and CaCl2 were 75 mmol·L -1, 0.5 mmol·L -1 and 5 mmol·L -1, respectively, Best effect. It is recommended to select the appropriate activation medium for artificial insemination in actual production.
