目的:以THP-1巨噬细胞源性泡沫细胞为研究对象,观察不同浓度黄芪多糖对胆固醇流出和ABCA1基因表达的影响。方法:将THP-1细胞诱导分化成泡沫细胞,用不同浓度黄芪多糖对其干预24h,γ计数仪检测胆固醇流出,RT-PCR及流式细胞仪检测ABCA1的表达。结果:黄芪多糖呈剂量依赖性(10-100mg/L)促进胆固醇流出;增加ABCA1的表达。结论:黄芪多糖可能通过增加ABCA1的表达而促进胆固醇流出。
OBJECTIVE: To investigate the effects of different concentrations of astragalus polysaccharides on cholesterol efflux and ABCA1 gene expression in THP-1 macrophage-derived foam cells. METHODS: THP-1 cells were induced to differentiate into foam cells and treated with different concentrations of astragalus polysaccharides for 24 hours. The cholesterol efflux was detected by gamma counter, and the expression of ABCA1 was detected by RT-PCR and flow cytometry. RESULTS: Astragalus polysaccharides promoted cholesterol efflux in a dose-dependent manner (10-100 mg/L); increased ABCA1 expression. Conclusion: Astragalus polysaccharides may promote cholesterol efflux by increasing ABCA1 expression.
