目的:观察高甘油三酯血症(HTG)状态下高密度脂蛋白(HDL)相关抗氧化酶——对氧磷酯酶-1(PON-1)的活性变化。方法:收集湖北武汉地区40例单纯性HTG患者和65例正常人群的血样,比较血脂分布特征,利用酶水解底物对氧磷的原理测定血浆HDL特征性PON-1的活性;同时以脂蛋白脂肪酶基因敲除(LPL-/-)鼠为HTG动物模型,检测血清中甘油三酯(TG)、总胆固醇(TC)含量和PON-1的活性改变。结果:①HTG组与对照组相比,血浆载脂蛋白(apo)B100水平显著升高(P<0.01),HDL-C、apoA-Ⅰ水平降低(P<0.05);PON-1酶活性显著降低(P<0.001);②HTG合并正常HDL-C组与低HDL-C对照组比较,PON-1活性无明显差别,但PON-1/apoA-Ⅰ比值显著降低(P<0.05);③LPL-/-鼠与对照组比较TG明显升高(P<0.001),PON-1活性显著下降(P<0.001)。结论:血清PON-1活性降低反映HDL抗氧化功能的异常,这可能与HTG易致动脉粥样硬化的发生密切相关。
Objective: To observe the changes of high density lipoprotein (HDL) -related antioxidant enzyme-paraoxonase-1 (PON-1) in hypertriglyceridaemia (HTG). Methods: Blood samples were collected from 40 patients with HTG and 65 normal controls in Wuhan, Hubei Province. Blood lipid profiles were compared. Lipoxygen was used to determine the activity of PON-1 in plasma. Lipase gene knockout (LPL - / -) mice were used as animal models of HTG. Serum levels of triglyceride (TG), total cholesterol (TC) and activity of PON-1 were detected. Results: ① Compared with the control group, the levels of apo B100, HDL-C and apoA-Ⅰ in the HTG group were significantly decreased (P <0.01), PON-1 activity was significantly decreased (P <0.001). (2) Compared with low HDL-C control group, there was no significant difference in PON-1 activity between HTG with normal HDL-C group and PON-1 / Compared with the control group, TG was significantly increased (P <0.001) and PON-1 activity was significantly decreased (P <0.001). Conclusion: The decrease of serum PON-1 activity reflects the abnormality of anti-oxidation function of HDL, which may be closely related to the development of HTG-induced atherosclerosis.
