[目的]观察氟染毒对体外培养新生大鼠皮肤成纤维细胞硬骨素(sclerostin,SOST)和Dickkopf-1(DKK1)蛋白表达的影响。[方法]采用原代培养方法,将大鼠皮肤成纤维细胞分为对照组(0 mg/L)和7个染氟剂量组(0.000 1、0.001、0.01、0.1、1、10、20 mg/L),分别在4个染氟时间段(24、48、72、96 h)收集细胞培养上清液,应用ELISA法测定SOST和DKK1蛋白表达水平。[结果]与染氟0 mg/L组相比,SOST表达量在染氟48 h的0.000 1、0.001、0.01 mg/L组,染氟72 h的1、10 mg/L组及染氟96 h的0.001、0.01、0.1、1 mg/L组均明显下降(P<0.05)。与染氟24 h组相比,SOST表达量在染氟48 h的0.01 mg/L组,染氟96 h的0.001、0.01、0.1、1、10 mg/L组均明显下降(P<0.05)。与染氟0 mg/L组相比,DKK1表达量在染氟48 h的0.001 mg/L组,染氟72 h的0.01、0.1、1、10、20 mg/L组及染氟96 h的0.01、0.1、1、10、20 mg/L组均明显下降(P<0.05)。与染氟24 h组相比,DKK1表达量在染氟48 h的0.000 1、0.001、0.01 mg/L组,染氟72 h的0.001、0.01、1、10、20 mg/L及染氟96 h的0.01、0.1、1、20 mg/L组均明显下降(P<0.05)。[结论]氟可导致大鼠皮肤成纤维细胞SOST和DKK1蛋白表达下调。
[Objective] To observe the effect of fluoride exposure on the expression of sclerostin (SOST) and Dickkopf-1 (DKK1) protein in cultured neonatal rat skin fibroblasts. [Method] The primary cultured rat skin fibroblasts were divided into control group (0 mg / L) and fluoride-treated group (0.000 1, 0.001, 0.01, 0.1, 1, L). Cell culture supernatants were harvested at 4, 4, 8, 72, 96 h, respectively. The expression of SOST and DKK1 protein was detected by ELISA. [Results] Compared with the 0 mg / L fluoride-exposed group, the expression of SOST was significantly lower in the 1, 10 mg / L group treated with fluoride at 1, 0, 10 and 0.01 mg / L for 48 h, h 0.001, 0.01, 0.1, 1 mg / L group were significantly decreased (P <0.05). Compared with the 24 h group, the expression of SOST was significantly decreased in the 0.01, 0.01, 0.1, 1 and 10 mg / L groups exposed to fluorine for 48 h and 96 h with fluorine (P <0.05) . Compared with the 0 mg / L fluoride-exposed group, the expression of DKK1 was significantly increased in the 0.001 mg / L group exposed to fluoride for 48 h, 0.01, 0.1, 1, 10, 20 mg / L for 72 h, 0.01,0.1,1,10,20 mg / L group were significantly decreased (P <0.05). Compared with 24 h group, the expression of DKK1 in the groups of 0.000 1, 0.001 and 0.01 mg / L for 48 h, 0.001, 0.01, 1, 10 and 20 mg / L for 72 h, h 0.01,0.1,1,20 mg / L group were significantly decreased (P <0.05). [Conclusion] Fluoride can decrease the expression of SOST and DKK1 protein in rat skin fibroblasts.
