目的探讨高效液相色谱柱前衍生法测定血、房水、晶状体中牛磺酸含量的价值。方法采用2种流动相,流动相A为0.05mol/L醋酸钠缓冲液(pH=6.5)甲醇四氢呋喃(80∶20∶0.8),流动相B为0.05mol/L柠檬酸缓冲液(pH=6.5)甲醇(20∶80);色谱柱为necleosODSΦ4.6nm×150nm。Wistar大鼠的血浆、房水、晶状体经处理和衍生后进样分析。结果该方法最低检出限为10μg/ml,氨基酸浓度与相对响应值之间平均相关系数为0.998±0.002,回收率为91.07%~96.79%,牛磺酸峰面积变异系数在进样量3μl时为10.75%,进样量5μl时为3.12%。结论高效液相色谱柱前衍生法可使血、房水、晶状体中的牛磺酸与其它氨基酸很好地分离,是测定牛磺酸含量的好方法。
Objective To investigate the value of pre-column derivatization of high performance liquid chromatography in the determination of taurine in blood, aqueous humor and lens. Methods Two kinds of mobile phases were used, the mobile phase A was 0.05 mol / L sodium acetate buffer (pH = 6.5) methanol tetrahydrofuran (80:20:0.8), the mobile phase B was 0.05 mol / L Citrate buffer (pH = 6.5) methanol (20:80); column for necleosODSΦ4.6nm × 150nm. Wistar rats plasma, aqueous humor, lens were processed and derived injection analysis. Results The detection limit of this method was 10μg / ml. The average correlation coefficient between amino acid concentration and relative response was 0.998 ± 0.002, the recovery was 91.07% ~ 96.79%. The peak area of taurine The coefficient of variation was 10.75% for a 3μl injection volume and 3.12% for a 5μl injection volume. Conclusion The pre-column derivatization method of HPLC can well separate the taurine from other amino acids in blood, aqueous humor and lens, which is a good method for the determination of taurine content.
