目的 :测定最新研制的注射用胰岛素缓释纳米囊中 (INC NC)胰岛素的含量。方法 :采用一阶导数紫外光谱法 ,并与高效液相色谱法 (HPLC)进行了比较。结果 :一阶导数紫外光谱法对该制剂低温离心所得上清液及沉淀中胰岛素含量测定的回收率分别为 1 0 0 31 %± 2 2 2 %和 1 0 0 66 %± 3 2 9% ;HPLC的回收率分别为 97 96 %±0 62 %和 96 34 %± 0 55 %。结论 :一阶导数紫外光谱法可快速、准确和简便地测定INS NC中INS的含量
OBJECTIVE: To determine the content of insulin in newly developed injectable insulin sustained-release nanocapsules (INC NC). Methods: The first-order derivative UV spectroscopy was used and compared with high-performance liquid chromatography (HPLC). Results: The recoveries of the first supernatant and the determination of insulin in the precipitate by the first derivative UV spectrophotometry were 1001% ± 222% and 1006% ± 392%, respectively. The HPLC recoveries were 97 96% ± 0 62% and 96 34% ± 0 55%, respectively. Conclusions: First-order derivative UV spectroscopy allows fast, accurate and easy determination of INS content in INS NC
