目的 动态观察灯盏细辛 (HEr)对金地鼠颊囊白斑癌变过程中α 平滑肌肌动蛋白 (α SMA)表达规律的影响 ,探讨HEr抗白斑癌变的可能机理。方法 金地鼠 10 2只随机分为模型组和HEr组 ,各用灯盏细辛流浸膏干预二甲基苯丙蒽 (DMBA)诱导的金地鼠白斑癌变进程。免疫组化技术检测涂布DMBA 4 9周的颊囊标本α SMA表达水平 ,并与模型组对照。结果 α SMA连续性染色血管密度平均值HEr组为 6 5 .76 ,模型组为 42 .12 (P <0 .0 0 1)。按10 0 %、5 0 %、2 0 %、10 %、3% 5级分档 ,HEr组的高分级例数远多于模型组 (P <0 .0 1)。结论 HEr有上调金地鼠白斑癌变过程中α SMA表达水平的确切作用 ,提示该药可能通过保持血管肌上皮细胞正常生理功能和血管基底膜完整性发挥抗癌变功效
Objective To observe the effect of Erigeron Breviscapus (HEr) on the expression of α smooth muscle actin (α SMA) during the carcinogenesis of gold hamster cheek pouch and to explore the possible mechanism of HEr anti-leukoplage carcinogenesis. Methods Two hundred gold hamsters were randomly divided into two groups: model group and HEr group. Each group was treated with Dengzhanxin extract to interfere with the progress of DMDM induced gold globus pallidus carcinogenesis. Immunohistochemical technique was used to detect the expression of α SMA in cheek pouch specimens coated with DMBA for 9 weeks and compared with the model group. Results The mean vascular density of α SMA staining was 65.76 in the HEr group and 42.12 in the model group (P < 0.01). According to the classification of 10%, 50%, 20%, 10%, and 3% of 5 grades, the number of high grade cases in the HEr group was far more than that of the model group (P < 0.01). Conclusion HEr can increase the expression of α SMA in leukoplakia carcinogenesis in gold hamsters, suggesting that this drug may play an anti-cancer effect by maintaining the normal physiological functions of vascular myoepithelial cells and the integrity of vascular basement membrane.
