目的 :通过肝素体外对涎腺腺样囊性癌肺高转移细胞株ACC M细胞与血小板粘附影响的实验 ,了解肝素抗ACC M肺转移的作用及其机制。方法 :健康成年人静脉血 ,制备富血小板血浆 (PRP) ,调整PRP血小板数至 2× 10 8/ml加入 1mMCaCl2 、1mMMgCl2 ,加入 2 4孔细胞培养板中 ,按不同分组要求在不同时间加入相应剂量的肝素后 3 7℃孵育 ,在不同时间消化贴壁粘附细胞 ,用 1%多聚甲醛固定后 ,用计数板进行细胞计数 ,计算粘附率。结果 :肝素体外对涎腺腺样囊性癌肺高转移细胞株ACC M细胞与血小板粘附有明显抑制作用。结论 :肝素体外对涎腺腺样囊性癌肺高转移细胞株ACC M细胞与血小板粘附有明显抑制作用 ,其机制可能是肝素通过与肿瘤细胞P 选择素配体的竞争作用抑制了P 选择素中介的血小板对肿瘤细胞的包被。
OBJECTIVE: To investigate the effect of heparin on ACC M lung metastasis and its mechanism by using heparin on ACC M cells and platelet adhesion in salivary gland adenoid cystic carcinoma cell line in vitro. Methods: Venous blood from healthy adults was used to prepare platelet-rich plasma (PRP). The number of PRP platelets was adjusted to 2 × 10 8 / ml by adding 1 mM CaCl 2 and 1 mM MgCl 2 into a 24-well cell culture plate. CONCLUSION: Heparin inhibits the adhesion of ACC M cells to salivary adenoid cystic carcinoma cell line ACC M in vitro. The possible mechanism is that heparin inhibits P-selectin by competition with P-selectin ligand of tumor cells Vegetarian platelets are coated on tumor cells.
