目的建立大鼠尿白蛋白放免测定方法及其应用意义。方法用大鼠白蛋白免疫青紫蓝兔,制备抗血清,采用双抗加PEG法。结果本方法测定范围25~1600μg/L,灵敏度为9.78μg/L,回收率89.2%~112.0%,批内、批间平均变异系数分别为4.1%和8.3%。大鼠在糖尿病成模2周时,24小时尿白蛋白排泄率(UAE)升高,而培哚普利治疗组尿白蛋白与正常大鼠组无明显差异,在治疗6个月后尿白蛋白明显低于糖尿病(P<0.01),培哚普利能明显抑制糖尿病大鼠尿白蛋白排泄,对糖尿病大鼠肾脏有某种保护作用。结论大鼠尿白蛋白放免测定方法的建立为糖尿病大鼠肾病模型和其他肾病的研究工作提供了一个客观的、灵敏的早期诊断指标
Objective To establish a method for determination of urinary albumin by radioimmunoassay in rats and its significance. Methods Qingzao rabbits were immunized with rat albumin to prepare antiserum. Double-antibody plus PEG method was used. Results The detection range of this method was 25 ~ 1600 μg / L with the sensitivity of 9.78 μg / L and the recovery of 89.2% ~ 112.0%. The average coefficient of variation (CV) was 4.1% and 8.3% . Urine albumin excretion rate (UAE) increased 24 hours after 2 weeks of diabetes mellitus in rats, while urinary albumin in perindopril group had no significant difference compared with normal rats. After 6 months of treatment, urinary white (P <0.01). Perindopril can significantly inhibit the urinary albumin excretion in diabetic rats, which may have some protective effects on the kidney of diabetic rats. Conclusion The establishment of rat urine albumin radioimmunoassay provides an objective and sensitive early diagnosis index for the research of nephropathy model and other nephropathy in diabetic rats
