以没食子酸为标样,研究Folin-Ciocalteu法测定未成熟黄瓜中总酚含量的最适测定条件。结果表明多酚含量的最佳测定条件为:未成熟黄瓜汁1 m L、10%Folin-Ciocalteu试剂5 m L、7.5%的Na2CO3溶液4 m L、常温反应60min、在765 nm波长下测定吸光度。多酚质量浓度在0~5μg/m L范围内与吸光度有很好的线性关系(Y=0.100 47X+0.021 53,相关系数r=0.998)。方法最低检出限为0.035μg/m L,定量限为0.175μg/m L,测定方法的平均回收率为100.49%,RSD为1.38%(n=6)。该方法的精密度高、稳定性好,适用于未成熟黄瓜中总酚含量的测定。
Using gallic acid as standard sample, the optimum conditions for the determination of total phenols in immature cucumber by Folin-Ciocalteu method were studied. The results showed that the optimum conditions for the determination of polyphenol content were as follows: 1 m L of unripe cucumber juice, 5 m L of 10% Folin-Ciocalteu reagent and 4 m L of 7.5% Na2CO3 solution, reacted at room temperature for 60 minutes and measured absorbance at 765 nm . The concentration of polyphenols in the range of 0 ~ 5μg / m L had a good linear relationship with absorbance (Y = 0.100 47X + 0.021 53, correlation coefficient r = 0.998). The method has the lowest detection limit of 0.035μg / mL and the limit of quantification of 0.175μg / mL. The average recovery of the method was 100.49% with RSD of 1.38% (n = 6). The method has high precision and good stability, and is suitable for the determination of the total phenol content in immature cucumber.
