目的:研究荷叶生物碱提取物对3T3-L1前脂肪细胞增殖分化的影响及其可能的作用机理。方法:采用MTT法检测荷叶生物碱提取物(LLAE)对前脂肪细胞生长的抑制效果并确定半抑制浓度(IC50);流式细胞仪技术测定细胞周期及凋亡情况;使用诱导剂对3T3-L1前脂肪细胞进行诱导分化,油红O染色法分析荷叶生物碱提取物(LLAE)对前脂肪细胞分化程度的影响。结果:荷叶生物碱提取物(LLAE)能显著地抑制3T3-L1前脂肪细胞增殖且呈剂量时间依赖性;在G0/G1期产生阻滞,有效诱导细胞凋亡;能显著地抑制3T3-L1前脂肪细胞分化,降低细胞中脂肪滴的积累。结论:荷叶生物碱提取物(LLAE)能有效地抑制3T3-L1前脂肪细胞的增殖及分化,揭示其可能具有预防肥胖的功效。
Objective: To study the effects of alkaloids from lotus leaf on the proliferation and differentiation of 3T3-L1 preadipocytes and its possible mechanism. Methods: MTT assay was used to determine the inhibitory effect of LLAE on the growth of preadipocytes and the half-inhibitory concentration (IC50). Flow cytometry was used to determine the cell cycle and apoptosis. The induction of 3T3 -L1 preadipocytes were induced to differentiate. Oil red O staining was used to analyze the effect of LLAE on the differentiation of preadipocytes. Results: The lotus leaf alkaloids extract (LLAE) could significantly inhibit the proliferation of 3T3-L1 preadipocytes in a time-and dose-dependent manner; block in G0 / G1 phase, effectively induced apoptosis; significantly inhibited 3T3- L1 preadipocytes differentiate, reducing the accumulation of lipid droplets in cells. Conclusion: The lotus leaf alkaloids extract (LLAE) can effectively inhibit the proliferation and differentiation of 3T3-L1 preadipocytes, revealing that it may have the effect of preventing obesity.
