目的研究H1N1流感病毒感染能力及机体免疫应答反应。方法利用0.1LD50剂量A/PR/8/34(H1N1)病毒感染C57BL/6小鼠,利用ELISA和流式细胞术等方法检测IL-5、IL-6水平和CD4+效应性T淋巴细胞比例。结果3dpi小鼠肺脏病毒复制达到高峰,为107EID50/g,7dpi肺脏组织病理学观察呈间质性肺炎、炎性细胞浸润;5dpi时IL-6水平达到峰值,为122pg/m L;7dpi时IL-5达到峰值,为680pg/m L;与未感染组相比,感染组CD4+CD44+CD62L-效应T细胞比例显著升高。结论结果表明PR8流感病毒感染小鼠,肺脏病毒复制水平高,机体免疫应答强,适合用于进一步的免疫学研究。
Objective To study the infection ability of H1N1 influenza virus and its immune response. Methods C57BL / 6 mice were infected with 0.1LD50 A / PR / 8/34 (H1N1) virus, and the levels of IL-5, IL-6 and CD4 + effector T lymphocytes were detected by ELISA and flow cytometry. Results The level of IL-6 in 3dpi mice peaked at 107EID50 / g for 7dpi, interstitial pneumonia and inflammatory cell infiltration at 7dpi, and reached 122pg / m L at 5dpi. -5 peaked at 680 pg / m L; the proportion of CD4 + CD44 + CD62L-effector T cells in the infected group was significantly higher than that in the uninfected group. Conclusion The results showed that PR8 influenza virus infection in mice, lung virus replication level, strong immune response, suitable for further immunological studies.
