十二氟-2-甲基-3-戊酮的急性吸入毒性试验

目的进行新型灭火剂十二氟-2-甲基-3-戊酮(C_6F_(12)O)气态化合物对小鼠的急性吸入毒性试验,为其使用时的安全性评价提供基础数据。方法将50只5周龄清洁级健康ICR种小鼠,体质量18~22 g,按数字表法随机分为5组,每组10只,雌雄各半,分别暴露于10%、15%、20%、25%、30%浓度下的灭火剂气体中2 h,采用静态吸入染毒法,用气相色谱仪监测染毒柜内化合物的浓度。观察小鼠的一般表现、中毒症状和死亡情况,试验结束后对死亡小鼠的肺部组织进行病理检查。结果随染毒浓度升高小鼠在实验期间出现死亡,C_6F_(12)O灭火剂对小鼠的急性吸入半数致死浓度(LC_(50))为3 648.5 g/m~3。病理检查显示死亡小鼠出现肺间质水肿。结论 C_6F_(12)O灭火剂的LC_(50)大于50 g/m~3,属于低等毒性,在小鼠急性吸入毒性试验中表现出较高的安全性。 OBJECTIVE To study the acute inhalation toxicity of gaseous compound of dodecafluoro-2-methyl-3-pentanone (C_6F_ (12) O) in mice, and to provide the basic data for the safety assessment of its use. Methods Fifty healthy 5-week-old healthy ICR mice were randomly divided into 5 groups (n = 10, n = 10) with 10 ~ 15% 20%, 25%, 30% concentration of the fire extinguishing agent gas 2 h, using static inhalation exposure method, using gas chromatography to monitor the concentration of the compound in the cabinet. Observe the general performance of mice, symptoms of poisoning and death, the pathological examination of lung tissue in the dead mice after the test. Results The death of mice was observed during the experimental period with the increase of exposure concentration. The acute inhalation concentration (LC_ (50)) of C_6F_ (12) O extinguishing agent in mice was 3 648.5 g / m ~ 3. Pathological examination showed dead mice with interstitial lung edema. Conclusion LC_ (50) of C_6F_ (12) O is more than 50 g / m ~ 3, which is of low toxicity. It shows high safety in mice acute inhalation toxicity test.
