目的:探讨中药肿节风溶液对人前列腺癌DU-145细胞增殖和凋亡的影响。方法:采用细胞培养的方法,用不同浓度的肿节风溶液作用于DU-145细胞48h,用MTT法检测用药后细胞的增殖情况,用流式细胞技术检测细胞的周期和凋亡状况。结果:肿节风溶液作用48h后,DU-145细胞表现形态和数量改变,出现数量减少,形态不规则,不同程度的细胞皱缩,边缘毛糙,染色质凝集或细胞核固缩碎裂;细胞增殖受到抑制,细胞周期阻滞于G2/M期,细胞凋亡率明显增加并呈一定的量效关系。结论:肿节风溶液促使人前列腺癌DU-145细胞形态改变,增殖抑制,细胞周期阻滞和加速细胞凋亡。 Objective: To investigate the effect of traditional Chinese medicine swollen wind solution on the proliferation and apoptosis of human prostate cancer DU-145 cells. Methods: DU-145 cells were treated with different concentrations of swollen cells for 48 hours by cell culture method. The proliferation of DU-145 cells was detected by MTT assay. The cell cycle and apoptosis were detected by flow cytometry. Results: After 48h, the number and morphology of DU-145 cells changed, the numbers of DU-145 cells changed, the number of DU-145 cells decreased, the morphology was irregular and the cells were shrinked with different degrees, rough edges, chromatin condensation or pyknosis; cell proliferation Inhibited, cell cycle arrest in G2 / M phase, the apoptosis rate was significantly increased and showed a certain dose-effect relationship. Conclusion: The solution of swollen lymph node promotes the morphological changes, proliferation inhibition, cell cycle arrest and accelerates apoptosis of human prostate cancer DU-145 cells.
