[目的]研究开胃进食汤超微配方颗粒对脾虚大鼠胃肠平滑肌细胞钙调蛋白(CaM)mRNA表达的影响,探求其对大鼠胃肠动力调节作用的可能机制。[方法]采用利血平致脾虚模型,大鼠随机分为正常组、模型组、开胃进食汤超微配方颗粒(超微)组、传统汤剂(传统)组和健胃消食片对照(健胃)组,用逆转录-聚合酶链反应(RT-PCR)半定量分析,观察各组CaM mRNA表达情况。[结果]模型组大鼠胃窦平滑肌内CaM mRNA表达明显减少,空肠平滑肌内明显增强(P<0.05);健胃组、传统组和超微组胃窦平滑肌内CaM mRNA表达较模型组增强(P<0.05),传统组和超微组空肠平滑肌内CaM mRNA表达低于模型组(P<0.05,<0.01)。[结论]开胃进食汤超微配方颗粒能促进胃窦平滑肌收缩和空肠平滑肌舒张,可能与其能明显提高胃窦平滑肌组织内和降低空肠平滑肌组织内CaM mRNA水平表达有关。
[Objective] To study the effect of appetizer-decoction Tangchao Formula Granules on the expression of calmodulin (CaM) mRNA in gastrointestinal smooth muscle cells of rats with spleen deficiency, and to explore its possible mechanism of gastrointestinal motility. [Methods] The model of spleen deficiency caused by reserpine was used. Rats were randomly divided into normal group, model group, appetite-fed soup ultra-micro-formulation granules (ultrasound) group, traditional decoction (traditional) group and Jianwei Xiaoshi tablets control. In the stomach group, semi-quantitative analysis was performed by reverse transcription-polymerase chain reaction (RT-PCR) to observe the expression of CaM mRNA in each group. [Results] The expression of CaM mRNA in gastric smooth muscle of guinea pigs in the model group was significantly decreased (P<0.05). The expression of CaM mRNA in the gastric antrum smooth muscle of Jianwei group, traditional group and ultra-micro group was stronger than that of the model group (P<0.05). P<0.05), CaM mRNA expression in jejunum smooth muscle of the traditional group and ultra-small group was lower than that of the model group (P<0.05, <0.01). [Conclusion] The appetizer-decoction soup ultra-micro-granules can promote the contraction of smooth muscle of gastric antrum and the relaxation of jejunal smooth muscle, which may be related to its ability to significantly increase the expression of CaM mRNA in antral smooth muscle tissue and decrease the level of jejunum smooth muscle tissue.
