目的探讨益气养阴、消牴通络中药对早期糖尿病大鼠肾组织Podocin基因表达的影响。方法将SD大鼠随机分为6组:肾切组、模型组、厄贝沙坦组、中药低、中、高剂量组,按各组相应药物剂量灌胃。于第6周末检测各组大鼠24 h尿蛋白(UPro)量及肾功能水平;RT-PCR检测肾皮质Podocin表达水平。结果厄贝沙坦、中药低、中剂量组与模型组比较UPro量显著下降(P<0.05);各治疗组与模型组比较,肾功能明显改善(P<0.05),且Podocin表达显著增加(P<0.01)。结论益气养阴、消牴通络中药可能通过减少Podocin蛋白的丢失,使肾小球滤过膜的完整性得到一定程度的恢复而减轻蛋白尿,从而发挥其对糖尿病肾病大鼠的治疗作用。
Objective To investigate the effects of Qi, nourishing yin and eliminating 牴 Tongluo Chinese medicine on Podocin gene expression in kidney of early diabetic rats. Methods The SD rats were randomly divided into 6 groups: Nephrectomy group, model group, irbesartan group, low, medium and high dose Chinese medicine group, according to the corresponding drug dose in each group. At the end of 6th week, the amount of urinary protein (UPro) and renal function of rats in each group were detected 24 hrs. The expression of Podocin in renal cortex was detected by RT-PCR. Results Compared with model group, irbesartan and traditional Chinese medicine low and medium dose groups showed a significant decrease in UPro (P <0.05). Compared with model group, renal function was significantly improved (P <0.05) and Podocin expression was significantly increased P <0.01). Conclusion Yiqi Yangyin and Xiaoyaotongluo Chinese herbs may reduce the proteinuria by reducing the loss of Podocin protein and restoring the integrity of the glomerular filtration membrane to a certain extent, thus exerting its therapeutic effect on diabetic nephropathy rats .
