从骆驼蓬(PeganumharmalaL.)种子中提取骆驼蓬总碱(totalalkaloidofharmala,TAH)。实验证明TAH对6种体外培养人癌细胞有强力细胞毒作用,对3种杂种鼠移植性肿瘤和3种人癌裸鼠移植物有明显抑瘤作用,与顺铂或阿霉素有协同作用。用电子显微镜观察TAH对肿瘤细胞超微结构影响;用显微荧光光度法分析对细胞周期的影响;用同位素掺入法测定对肿瘤细胞大分子合成的影响:用自建高效液相法进行骆驼蓬碱代谢动力学研究;毒性研究表明长期大剂量TAH可引起可逆性肾毒性。
Total alkaloid (totalalkaloid of harala, TAH) was extracted from Peganum harmala L. seeds. Experiments show that TAH on six kinds of human cancer cells in vitro has a strong cytotoxic effect on the three kinds of hybrid mice transplanted tumors and three kinds of human cancer nude mice transplantation significantly inhibit the tumor and cisplatin or doxorubicin synergistic effect . The effect of TAH on the ultrastructure of tumor cells was observed by electron microscopy. The effect of TAH on the cell cycle was analyzed by fluorescence microscopy. The effect of TAH on the synthesis of tumor macrophages was determined by isotope incorporation method. Peng base metabolic kinetics studies; toxicity studies have shown that long-term high-dose TAH can cause reversible nephrotoxicity.
