目的研究27种常见中药材甲醇提取物对黄嘌呤氧化酶的抑制活性。方法以别嘌醇为阳性对照,采用全波长酶标仪测定反应体系的吸光度,测定波长为295 nm,通过Graph Pad Prism 5软件计算抑制率。结果 27种中药甲醇提取物均对黄嘌呤氧化酶显示不同程度的抑制作用,其中6种中药(丹参、野菊花、牡丹皮、川芎、当归、远志)对黄嘌呤氧化酶具有较好的抑制作用,其IC_(50)分别为91.2、39.5、87.7、11.8、54.6、94.1μg/ml(阳性别嘌醇的IC50为6.4μg/ml)。结论该研究首次发现牡丹皮、川芎和远志具有显著的黄嘌呤氧化酶抑制活性,此结果可为揭示这些中药的抗痛风作用提供科学数据,同时亦为其进一步开发利用提供科学参考。
Objective To study the inhibitory activity of methanol extracts of 27 common Chinese herbs on xanthine oxidase. Methods Allopurinol was used as a positive control. The absorbance of the reaction system was measured by a full-wavelength microplate reader at a wavelength of 295 nm. The inhibition rate was calculated by Graph Pad Prism 5 software. Results The results showed that all of the 27 Chinese herbal extracts had different inhibitory effects on xanthine oxidase, of which six kinds of traditional Chinese medicines (Salvia miltiorrhiza, Chrysanthemum morifolium, Cortex Moutan, Chuanxiong, Angelica and Polygalaceae) had a good inhibitory effect on xanthine oxidase , And their IC 50 values were 91.2, 39.5, 87.7, 11.8, 54.6 and 94.1 μg / ml, respectively (IC50 of positive allopurinol was 6.4 μg / ml). Conclusions This study, for the first time, found that Cortex Moutan, Chuanxiong Rhizoma and Polygalaceae have significant xanthine oxidase inhibitory activity. These results can provide scientific data for revealing the anti-gout effect of these traditional Chinese medicines and provide a scientific reference for their further development and utilization.
