目的探讨系统性红斑狼疮(SLE)患者外周血淋巴细胞Caspase-1和Caspase-3的表达水平及其与淋巴细胞凋亡的关系。方法应用逆转录-聚合酶链反应(RT-PCR)检测34例活动期SLE患者和30例正常人外周血单一核细胞(PBMC)中Caspase-1和Caspase-3mRNA的表达水平。结果活动期SLE患者PBMC中Caspase-1mRNA的平均表达水平(1.20±0.33)明显高于正常人对照组(0.79±0.25),差异有非常显著性(P<0.01);Caspase-3mRNA的平均表达水平(0.87±0.16)高于正常人对照组(0.66±0.10),差异有显著性(P<0.05)。活动期SLE患者PBMC中Caspase-1和Cas-pase-3的阳性表达率与正常人对照组相比差异无显著性(P>0.05)。结论活动期SLE患者外周血淋巴细胞Caspase-1和Caspase-3mRNA表达水平增高,Caspase级联反应可能参与了SLE患者淋巴细胞的凋亡过程。
Objective To investigate the expression of Caspase-1 and Caspase-3 in peripheral blood lymphocytes in patients with systemic lupus erythematosus (SLE) and its relationship with apoptosis of lymphocytes. Methods The mRNA expression of Caspase-1 and Caspase-3 in peripheral blood mononuclear cells (PBMCs) of 34 patients with active SLE and 30 normal controls were detected by reverse transcription-polymerase chain reaction (RT-PCR) Results The average expression level of Caspase-1 mRNA in PBMC of active SLE patients (1.20 ± 0.33) was significantly higher than that of normal controls (0.79 ± 0.25) (P <0.01). The average expression level of Caspase-3 mRNA (0.87 ± 0.16) was higher than that of normal control group (0.66 ± 0.10), the difference was significant (P <0.05). The positive rates of Caspase-1 and Caspase-3 in PBMC of active SLE patients were not significantly different from those of normal controls (P> 0.05). Conclusions The expression of Caspase-1 and Caspase-3 mRNA in peripheral blood lymphocytes of patients with active SLE is increased. Caspase cascade may play an important role in the apoptosis of lymphocytes in patients with SLE.
