目的研究金丝桃素对体外培养的人视网膜色素上皮细胞(RPE)凋亡的影响,探讨药物防止增生性玻璃体视网膜病变(PVR)的某些基本机理。方法用含0.5μM~5μM金丝桃素不同浓度的血清培养人RPE,流式细胞仪(FCM)检测调亡细胞定量。结果金丝桃素作用后RPE的S期百分比随着金丝桃素浓度增加而逐渐减少(金丝桃素4种浓度P均<0.01)。凋亡细胞定量结果显示:在2.5μM和5μM金丝桃素组凋亡细胞的比例分别为2.3%和6.1%。结论2.5μM和5.0μM金丝桃素能诱导RPE的凋亡,运用于眼内来控制RPE的前景是光明的。
Objective To study the effect of hypericin on the apoptosis of human retinal pigment epithelial cells (RPE) cultured in vitro, and to explore some basic mechanisms of drugs for preventing proliferative vitreoretinopathy (PVR). Methods Human RPE was cultured with different concentrations of serum containing 0.5μM-5μM hypericin, and apoptosis was measured by flow cytometry (FCM). Results The percentage of S-phase of RPE after hypericin treatment decreased with the increase of hypericin concentration (P <0.01 for hypericin). The quantitative results of apoptotic cells showed that the proportion of apoptotic cells in the 2.5 μM and 5 μM hypericin group was 2.3% and 6.1%, respectively. Conclusion 2.5μM and 5.0μM hypericin can induce apoptosis of RPE. The prospect of using RPE in the eye to control the eyes is bright.
