目的总结本研究中心的SD大鼠胚胎-胎仔发育毒性试验各个指标的正常值范围,建立SD大鼠胚胎-胎仔发育指标的背景数据,为生殖毒理研究提供参考。方法从本研究中心近期的生殖毒性试验中选取多批SD大鼠胚胎-胎仔发育毒性试验,整理对照组孕鼠的胚胎发育和胎仔生长发育指标数据,包括孕鼠的窝平均黄体数、着床数、平均活胎数、活胎率、吸收胎率、死胎率,胎仔体重、胎盘重、身长、尾长、外观畸形、内脏和骨骼发育指标等,将数据进行统计,分别计算各个指标的均数和标准差。结果本研究总结了214只孕鼠和2734只活胎仔的胚胎-胎仔发育指标。本研究所总结的孕鼠年龄约为50~90日龄,孕鼠妊娠第0天(GD 0)体重为(229±20)g,妊娠第20天(GD 20)体重为(380±34)g,整个孕期体重增重较大,食量随孕期增长而逐渐增加。GD 20安乐死并实施解剖,孕鼠窝平均黄体数为(15.6±2.8)个,窝平均着床数为(13.6±3.7)个,窝平均活胎数为(12.8±3.7)只,总吸收胎数为177(177/2734),早期吸收胎数为175(175/2734),晚期吸收胎数为2(2/2734),死胎数为0(0/2734),活胎率为(93.96±9.54)%,吸收胎率为(6.04±9.54)%,早期吸收胎率为(5.98±9.40)%,晚期吸收胎率为(0.07±0.69)%,死胎率为(0.00±0.00)%。胎仔发育指标中,雌性胎仔体重为(3.751±0.325)g,雄性胎仔体重为(3.942±0.364)g,胎仔胎盘重为(0.558±0.112)g,胎仔身长(37.17±1.41)mm,尾长为(13.17±0.79)mm,胎仔性别比为雄性/雌性=1291/1443=0.9。活胎仔中,胎仔外观畸形率为(0.06±0.64)%;骨骼变异胎仔率为(4.22±13.24)%,骨骼异常胎仔率为(2.27±8.17)%;内脏变异胎仔率为(25.81±25.51)%,内脏异常胎仔率为(0.88±4.43)%。结论本中心的SD大鼠胚胎-胎仔发育毒性试验各个指标的改变均比较稳定,胎仔自发畸形率较低。
OBJECTIVE: To summarize the normal range of various indicators of SD rat embryo-fetal developmental toxicity test and to establish the background data of SD rat embryo-fetal development index, so as to provide reference for reproductive toxicology research. Methods From the recent reproductive toxicity test in this research center, multiple batches of embryonic-fetal developmental toxicity test of SD rats were selected and the data of embryo development and fetal growth and development of pregnant rats in the control group were collected. The average litter size, The average number of live births, live birth rate, abortion rate, stillbirth rate, birth weight, placental weight, body length, tail length, appearance deformity, visceral and skeletal development indicators, the data were calculated, were calculated for each indicator Number and standard deviation. Results The present study summarized the embryo-fetal development of 214 pregnant and 2734 live fetuses. The pregnant rats of this study are about 50-90 days old. The weight of pregnant rats on day 0 (GD 0) was (229 ± 20) g, and the weight on day 20 (GD 20) was (380 ± 34) g, weight gain throughout the larger pregnancy, food intake increased with the increase of pregnancy. GD 20 was euthanized and dissected. The average corpus luteum of pregnant rats was (15.6 ± 2.8), the average number of implantation was (13.6 ± 3.7), the average number of live litter was (12.8 ± 3.7) 177 (177/2734) in the early stage, 175 (175/2734) in the early stage, 2 (2/2734) in the late stage, 0 (0/2734) stillbirth and (93.96 ± 9.54%, the rate of the fetus was 6.04 ± 9.54%, the rate of the early fetus was 5.98 ± 9.40%, that of the late fetus was 0.07 ± 0.69%, and the stillbirth rate was 0.00 ± 0.00%. The average fetal weight was (3.751 ± 0.325) g, the male fetus weight was (3.942 ± 0.364) g, the fetal placenta weight was (0.558 ± 0.112) g, the fetal body length was (37.17 ± 1.41) mm, the tail length was (13.17 ± 0.79) mm, the sex ratio of the fetus was male / female = 1291/1443 = 0.9. In the live fetus, the fetal malformation rate was (0.06 ± 0.64)%; the skeletal variant fetus rate was (4.22 ± 13.24)%, the skeletal abnormal fetus rate was (2.27 ± 8.17)%; the visceral variation fetus rate was (25.81 ± 25.51) %, Visceral abnormal fetal rate was (0.88 ± 4.43)%. Conclusion The SD rat embryo - fetal developmental toxicity test of all indicators of changes are relatively stable, fetal spontaneous deformity rate is low.
