应用PCR技术从白纹伊蚊和埃及伊蚊基因组中扩增出defensin成熟肽基因并对其分子生物学特性进行分析 ,发现埃及伊蚊 (BoraBora株、海口株 )、白纹伊蚊 (成都株、宜兴株、广州株 )推导的氨基酸序列与埃及伊蚊defensinA相同 ;通过Chou Fasman预测法对defensinA成熟肽的二级结构进行预测 ,可以看出在defensin成熟肽氨基酸序列的 15~ 2 3位有一个α 螺旋 ,35~ 39位有一个 β 折叠 ,32~ 34位有一个转角 ;通过同源性比较推测蚊虫defnsinA的三级结构
The gene of defensin mature peptide was amplified from Aedes albopictus and Aedes aegypti by PCR and the molecular biological characteristics were analyzed. It was found that Aedes aegypti (Bora Bora strain, Haikou strain), Aedes albopictus (Chengdu strain , Yixing strain and Guangzhou strain) were the same as those of defensin A in Aedes aegypti. Prediction of the secondary structure of the mature peptide of defensin A by Chou Fasman prediction method showed that in the amino acid sequence of defensin mature peptide, An α-helix with a β-sheet at positions 35-39 and a turn at positions 32-34; deduced from the homology comparison of the tertiary structure of defensinA
