目的:探讨补肾填精方促进大鼠慢性难愈性创面修复愈合的作用及机制。方法:在52只雄性SD大鼠背部制造全层皮肤缺损开放性创面,除正常对照组外,其余大鼠肌肉注射醋酸氢化可的松建立难愈性创面模型,随机分为模型组、补肾填精方组,观察各组大鼠的创面愈合率、愈合时间;并通过免疫组化及图像分析等技术,观察各组大鼠创面肉芽组织中血管内皮生长因子(vascular endothelial growth factor,VEGF)表达情况和微血管计数(microvessel count,MVC)。结果:模型组与正常对照组比较,各时间点创面愈合率均显著降低(P<0.01,P<0.05),愈合时间明显延长(P<0.01);补肾填精方组与模型组比较,各时间点创面愈合率均显著提高(P<0.01),愈合时间明显缩短(P<0.01)。模型组的VEGF表达量和MVC均显著低于正常对照组(P<0.01);补肾填精方组与模型组比较,VEGF表达量、MVC均明显升高(P<0.01)。结论:补肾填精方能明显促进大鼠慢性难愈性创面修复愈合,其作用机制可能与上调创面VEGF表达,诱导血管新生有关。
Objective: To investigate the effect and mechanism of Bushen Tianjing Recipe in promoting healing of chronic refractory wounds in rats. METHODS: Full thickness skin defect open wounds were made on the back of 52 male Sprague-Dawley rats. Except for the normal control group, the other rats were intramuscularly injected with hydrocortisone acetate to establish a refractory wound model and were randomly divided into model group, The wound healing rate and healing time of rats in each group were observed. The expression of vascular endothelial growth factor (VEGF) in wound granulation tissue in each group was observed by immunohistochemistry and image analysis techniques Condition and microvessel count (MVC). Results: Compared with the normal control group, the wound healing rate at each time point was significantly lower (P <0.01, P <0.05) and the healing time was significantly longer (P <0.01). Compared with the model group, The wound healing rate at the time point was significantly increased (P <0.01), the healing time was significantly shorter (P <0.01). The expression of VEGF and MVC in the model group were significantly lower than those in the normal control group (P <0.01). Compared with the model group, the expression of VEGF and the MVC of the model group were significantly increased (P <0.01). Conclusion: Bushen Tianjing Recipe can significantly promote healing of chronic refractory wounds in rats. The mechanism may be related to up-regulation of VEGF expression and induction of angiogenesis in wounds.
