目的 :探索胰岛素样生长因子 -1(in sulin likegrowthfactor 1,IGF 1)及其受体 (in sulin likegrowthfactor 1receptor ,IGF 1R)在胃癌发生、发展中的作用 ,寻找新的胃癌早期诊断标志物 ,并为胃癌的病因研究提供理论依据。方法 :对 2 8例经手术切除并由病理证实的胃癌患者的癌、癌旁及胃黏膜正常区组织标本进行研究 ,以 14例正常人胃黏膜标本作对照 ,应用RT PCR方法 ,检测IGF 1/IGF 1R在胃癌组织与正常人胃黏膜中的表达变化和胃癌患者的癌、癌旁及胃黏膜正常区组织中的表达变化。结果 :IGF 1/IGF 1RmRNA的表达量在胃癌组明显高于正常对照组。在胃癌组织中IGF 1/β actin比值为 0 3 0 5± 0 0 3 2 ,IGF 1R/β actin比值为 0 12 9± 0 0 11;在正常胃黏膜中IGF 1/β actin比值为 0 15 4± 0 0 13 ,IGF 1R/β actin比值为 0 0 2 7± 0 0 0 4。两组间的差异有统计学意义 ,P <0 0 1;且mRNA的表达量按正常、癌旁、癌组织顺序逐渐增高 ,3组间差异有统计学意义 ,P <0 0 5。结论 :IGF 1/IGF 1R因子的高表达 ,促进了胃黏膜上皮细胞的异常增殖 ,在胃癌的发生发展中起重要的作用。
Objective: To explore the role of in sulin-like growth factor-1 (IGF-1) and its receptor (IGF-1R) in the development and progression of gastric cancer and to search for a new early diagnosis marker of gastric cancer. Provide a theoretical basis for the study of the etiology of gastric cancer. Methods: Tissues of 28 normal gastric mucosa and normal gastric mucosa of 28 gastric cancer patients with surgically resected resected pathology were studied, and 14 normal gastric mucosa specimens were used as control. RT-PCR was used to detect IGF 1 / IGF 1R expression in gastric mucosa and normal gastric mucosa and gastric cancer patients with cancer, paracancer and gastric mucosa in normal tissue changes. Results: The expression of IGF 1 / IGF 1 R mRNA in gastric cancer group was significantly higher than that in normal control group. The ratio of IGF 1 / β actin in gastric cancer tissue was 0 30 05 ± 0 0 3 2, the ratio of IGF 1R / β actin was 0 12 9 ± 0 0 11, and the ratio of IGF 1 / β actin in normal gastric mucosa was 0 15 4 ± 0 0 13, IGF 1R / β actin ratio was 0 0 2 7 ± 0 0 0 4. The difference between the two groups was statistically significant, P <0 01; and the expression of mRNA by normal, paracancerous, cancer order gradually increased, the difference between the three groups was statistically significant, P <0 05. Conclusion: The high expression of IGF-1 / IGF-1R promotes the abnormal proliferation of gastric mucosal epithelial cells and plays an important role in the development of gastric cancer.
