目的探讨同种异基因大鼠骨髓间充质干细胞(MSCs)联合骨髓细胞(BMCs)输注对胰岛移植大鼠体内细胞因子水平的影响。方法制备SD大鼠DM模型,Wistar大鼠骨髓传代纯化获得MSCs。经静脉输注供体MSCs和/或BNCs后7d,糖尿病SD大鼠进行同种异体胰岛移植,观测胰岛移植物存活时间,检测白介素-2(IL-2)、干扰素-γ(IFN-γ)、白介素-4(IL-4)、白介素-10(IL-10)、白介素-6(IL-6)及转化生长因子-β1(TGF-β1)含量。结果预输注高浓度MCSs联合BMCs能显著延长移植胰岛的生存时间,平均为30d。该组大鼠体内IL-2、IFN-γ水平降低,IL-10和TGF-β1水平升高(P<0.05)。结论高浓度同种异基因大鼠MSCs联合BMCs输入体内后,使胰岛移植大鼠血清细胞因子发生显著变化,可能是诱导胰岛移植免疫耐受的原因之一。
Objective To investigate the effect of allogeneic rat bone marrow mesenchymal stem cells (MSCs) combined with bone marrow cells (BMCs) infusion on the level of cytokines in islet transplantation rats. Methods SD rats were prepared DM model, Wistar rat bone marrow purification of MSCs. Allograft islets were transplanted into diabetic SD rats 7 days after intravenous infusion of donor MSCs and / or BNCs, the survival time of islet grafts was observed, and the levels of interleukin-2 (IL-2), interferon-γ (IL-4), interleukin-10 (IL-10), interleukin-6 (IL-6) and transforming growth factor-β1 (TGF- Results The pretreatment with high concentrations of MCSs combined with BMCs significantly prolonged the survival time of transplanted islets with an average of 30 days. The levels of IL-2 and IFN-γ in the rats were decreased and the levels of IL-10 and TGF-β1 were increased (P <0.05). CONCLUSIONS: MSCs combined with BMCs from high concentration allogeneic rats can significantly change serum cytokines in rats after islet transplantation, which may be one of the reasons for inducing immune tolerance of islet transplantation.
