目的通过检测高脂膳食8周后肥胖倾向、肥胖抵抗和对照组小鼠血清雄激素、雌激素和瘦素水平,探讨瘦素和性激素的关系以及与雄性生殖功能之间的关系。方法将SPF级雄性C57bl/6小鼠分为对照组、实验组,对照组给予基础饲料,实验组给予高脂饲料。饲养8周后根据实验组小鼠体重分为肥胖倾向组和肥胖抵抗组,每组10只。分离各组小鼠睾丸、附睾与精囊腺称重,电镜检测睾丸的病理改变,ELISA方法检测血清雄激素、雌激素和瘦素水平。结果高脂饮食8周后,肥胖倾向组小鼠睾丸脏器系数低于对照组,差异有统计学意义(P<0.05);肥胖倾向、肥胖抵抗组小鼠的精子活动率分别为(40.97±21.84)%,(42.78±8.22)%,均明显低于对照组[(66.17±17.8)%],差异有统计学意义(P<0.01);肥胖倾向、肥胖抵抗组小鼠的雄激素水平低于对照组,瘦素水平高于对照组,差异有统计学意义(P<0.05,P<0.01)。电镜观察发现,高脂饮食小鼠睾丸间质细胞可见大量脂肪细胞,核型不规则,核内异染色质边集。结论肥胖雄性小鼠瘦素水平升高和睾丸间质细胞受损可能与雄激素水平下降有关。
OBJECTIVE: To investigate the relationship between leptin and sex hormones and the relationship with male reproductive function by measuring the level of serum androgen, estrogen and leptin in obesity proneness, obesity resistance and control mice after 8 weeks of high-fat diet. Methods SPF male C57bl / 6 mice were divided into control group, experimental group and control group. The experimental group was given high fat diet. After eight weeks of feeding, mice in the experimental group were divided into obesity-prone group and obesity-resistant group according to the body weight of the experimental group, with 10 rats in each group. The testes, epididymis and seminal vesicles of each group were separated and weighed. The pathological changes of testis were detected by electron microscopy. The levels of serum androgen, estrogen and leptin were detected by ELISA. Results After 8 weeks of high-fat diet, the organ coefficient of testis in obesity prone group was lower than that in control group (P <0.05). The sperm motility of obesity prone group and obesity resistant group was (40.97 ± 21.84)% and (42.78 ± 8.22)% respectively, which were significantly lower than those in the control group (66.17 ± 17.8%) (P <0.01). The obesity tended to be lower in androgen-resistant mice In the control group, leptin level was higher than that of the control group, the difference was statistically significant (P <0.05, P <0.01). Electron microscopy showed that a large number of adipocytes were observed in testicular stromal cells in mice fed high-fat diet. The karyotypes were irregular and the heterochromatin margins in the nucleus were observed. Conclusion The increase of leptin level and the impairment of testicular interstitial cells in obese male mice may be related to the decrease of androgen levels.
