目的研究虎眼万年青总皂苷对人乳腺癌MDA-MB-231细胞增殖和凋亡的影响。方法用MTT比色法检测虎眼万年青总皂苷对MDA-MB-231细胞的增殖抑制作用;流式细胞术分析细胞凋亡与细胞周期变化;透射电镜观察细胞形态学变化。结果虎眼万年青总皂苷对MDA-MB-231细胞生长有明显抑制作用,并呈剂量、时间依赖性;可诱导细胞发生凋亡,且凋亡率呈浓度依赖性,使细胞周期明显阻滞于G0/G1期;电镜下可见典型的细胞凋亡形态学改变。结论虎眼万年青总皂苷可抑制人乳腺癌MDA-MB-231细胞增殖,并可诱导细胞凋亡,其机制可能与虎眼万年青总皂苷将癌细胞阻止于G0/G1期相关。
Objective To study the effects of Ornithogalum vulgare on proliferation and apoptosis of human breast cancer MDA-MB-231 cells. Methods MTT colorimetry was used to detect the proliferation of MDA-MB-231 cells. The apoptosis and cell cycle changes were analyzed by flow cytometry. The morphological changes of cells were observed by transmission electron microscope. Results The results showed that total saponin of Ornithogalum coronatum could significantly inhibit the growth of MDA-MB-231 cells in a dose-and time-dependent manner and induced apoptosis in a concentration-dependent manner, which significantly blocked the cell cycle G0 / G1 phase; typical apoptotic morphological changes were observed under electron microscope. Conclusion The total saponins of Ornithogalum vincaensis can inhibit the proliferation and induce the apoptosis of human breast cancer cell line MDA-MB-231, which may be related to the arrest of G0 / G1 phase cells by the total saponins of Ornithogalum.
