[目的] 研究归芪方对糖尿病(DM)大鼠早期肾脏病变的保护作用及其机制。[方法] Wistar大鼠腹腔注射链脲佐菌素建立DM模型,随机分为DM模型组(B组)、苯那普利治疗组(C组)、归芪方治疗组(D组),另设正常对照组(A组)。实验第8周时,观察各组血糖、血脂、肾重/体重及蛋白尿的改变,放射免疫法测定血浆及肾组织血管紧张素Ⅱ(AngⅡ),半定量RT PCR法检测肾脏转化生长因子 β1(TGF β1)mRNA的表达。[结果] 归芪方可以降低高血糖、高血脂,并且在改善肾脏肥大及抑制肾脏 TGF β1 mRNA 表达方面优于苯那普利(P < 0.01, <0.05),归芪方还能降低肾内和血浆AngⅡ水平(P<0.01)。[结论] 归芪方可调节糖、脂代谢紊乱,降低 AngⅡ水平,抑制肾脏TGF β1 mRNA的表达,改善肾功能,延缓DM的进展。
[Objective] To study the protective effect and mechanism of Guiqi decoction on early renal lesion in diabetic rats. [Methods] Wistar rats were intraperitoneally injected with streptozotocin to establish DM models and were randomly divided into DM model group (B group), Benazepril treatment group (C group), and Gui Qi Fang treatment group (D group). Let normal control group (group A). At the 8th week of the experiment, blood glucose, blood lipids, kidney weight/body weight and proteinuria were observed in each group. Plasma and renal angiotensin II (Ang II) were measured by radioimmunoassay. Transforming growth factor β1 was measured by semi-quantitative RT PCR. (TGFβ1) mRNA expression. [Results] Guizhi decoction can reduce hyperglycemia and hyperlipidemia, and it is superior to benazepril in improving renal hypertrophy and inhibiting TGF-β1 mRNA expression in kidney (P < 0.01, <0.05). And plasma Ang II levels (P <0.01). [Conclusion] Guiqifang can regulate glucose, lipid metabolism, reduce the level of AngII, inhibit the expression of TGFβ1 mRNA in kidney, improve renal function and delay the progression of DM.
