目的:探讨白细胞介素1β(IL1β)在子痫前期患者胎盘中的表达及缺氧对胎盘绒毛产生IL1β的影响。方法:采用半定量PCR方法分别检测子痫前期患者及正常孕妇2组各6例胎盘IL1βmRNA水平;并在缺氧(20mL/L)及常氧(210mL/L)环境下培养8例正常足月胎盘绒毛,通过ELISA检测胎盘绒毛培养液中IL1β的含量。结果:半定量PCR结果显示IL1β在子痫前期患者胎盘中mRNA水平明显高于正常足月妊娠组(P<0.05);在缺氧条件下胎盘绒毛培养上清中IL1β的含量非常显著高于常氧条件下(P<0.01)。结论:IL1βmRNA在子痫前期患者胎盘中高表达,提示IL1β可能参与了妊娠期高血压疾病的病理生理过程;胎盘缺氧可能是妊娠期高血压疾病患者胎盘IL1β水平增高的一个重要因素。
Objective: To investigate the expression of interleukin 1β (IL1β) in placenta of preeclampsia and the effect of hypoxia on the production of IL1β in placenta. Methods: The levels of IL1βmRNA in placenta of 6 patients with preeclampsia and 2 normal pregnant women were detected by semi-quantitative PCR. Eight normal pregnant subjects were cultured in hypoxia (20 mL / L) and normoxia (210 mL / L) Placental villi, IL1β content in placental villi culture solution was detected by ELISA. Results: The results of semi-quantitative PCR showed that the level of IL1β mRNA in placenta of preeclampsia was significantly higher than that of normal term pregnancy group (P <0.05). The content of IL1β in placental villi culture supernatant under hypoxia was significantly higher than that of normal Oxygen conditions (P <0.01). CONCLUSION: IL1βmRNA is highly expressed in the placenta of patients with preeclampsia, suggesting that IL1β may be involved in the pathophysiological process of hypertensive disorder complicating pregnancy. Placental hypoxia may be an important factor in the increase of placental IL1β level in patients with gestational hypertension.
