IL-12、IL-7、IL-2体外协同诱导人PBMC增殖和PBMC杀瘤活性的实验研究

研究了重组IL12、IL7、IL2协同对健康人外周血单个核细胞(PBMC)的体外增殖和杀瘤活性的影响。结果表明,高浓度IL2与低浓度IL2协同可引起PBMC明显增殖,若在该培养体系中加入低浓度的IL2,可使这种增殖效应显著增强。如果加大IL12剂量时会产生更强的细胞毒活性,并呈现剂量依赖关系。在此培养条件下的培养上清中亦检出了较高水平的IFNγ。对培养3~14d的协同刺激细胞表型分析显示,CD56抗原阳性细胞百分率呈递增趋势,提示CD56+的NK细胞构成了产生增殖和杀伤效应的主要细胞群体。 To study the recombinant IL  12, IL  7, IL  2 synergistic effect on human peripheral blood mononuclear cells (PBMC) in vitro proliferation and tumorigenicity. The results showed that high concentrations of IL 2 and low concentrations of IL 2 synergistic PBMC can cause significant proliferation, if the culture system by adding a low concentration of IL 2, can make such a proliferation effect was significantly enhanced. If you increase the IL 12 dose will produce more cytotoxic activity, and showed a dose-dependent relationship. In this culture conditions, the culture supernatant also detected a high level of IFN γ. Analysis of the co-stimulatory cell phenotypes after 3 to 14 days of culture showed that the percentage of CD56 antigen positive cells showed an increasing trend, suggesting that CD56 + NK cells constitute the major cell population that produces proliferative and cytotoxic effects.
