目的:本研究以家兔为研究对象,探讨丹参静脉给药后其水溶性标志成份丹参素(danshensu,Dhpl)能否通过血-眼屏障到达靶组织直接发挥作用,试图阐述静脉注射中药治疗眼病的作用方式及眼内的代谢,为临床提供合理的用药方案。方法:利用高效液相色谱法(HPLC)测定Dhpl在血液、房水、玻璃体内的浓度。结果:在此色谱条件下,Dhpl的保留时间为6.776分钟,无其他峰干扰,方法准确度和精密度符合生物样品的测定要求。结论:本方法灵敏度、准确度高,特异性强,可用于血液、房水、玻璃体中Dhpl质量浓度测定。
Objective: In this study, rabbits as the research object, to explore Salvia intravenous administration of its water-soluble flag ingredient Danshensu (danshensu, Dhpl) can reach the target tissue directly through the blood-eye barrier to play a direct role, trying to elaborate intravenous Chinese medicine treatment of eye disease The mode of action and the metabolism of the eye, to provide a reasonable clinical medication program. Methods: The concentration of Dhpl in blood, aqueous humor and vitreous was determined by high performance liquid chromatography (HPLC). Results: Under this chromatographic condition, the retention time of Dhpl was 6.776 minutes and no other peak interference was found. The accuracy and precision of the method met the requirements of biological samples. Conclusion: The method is sensitive, accurate and specific and can be used for the determination of Dhpl concentration in blood, aqueous humor and vitreous.
