目的 筛选与受损内皮细胞特异性结合的短肽。方法 以氧化型低密度脂蛋白 (oxidizedlowdensitylipoprotein ,ox -LDL)损伤牛胸主动脉内皮细胞 ,用丝状噬菌体 (FUSE5 )表面展示的随机 6肽和 15肽文库进行混合筛选 ,通过DNA序列测定确定其插入的氨基酸序列。结果 经过 3轮筛选 ,得到一些特异性结合的噬菌体克隆 ,随机挑选 10个克隆 ,进行DNA序列分析 ,其中 8个克隆含 6肽序列 ,2个克隆含 15肽序列。结论 用噬菌体技术能筛到与受损内皮细胞亲和力高、结合力强的特异性短肽
Objective To screen short peptides that specifically bind to damaged endothelial cells. Methods Oxidized low density lipoprotein (ox-LDL) was used to injure the bovine thoracic aorta endothelial cells. The random 6-peptide and 15-peptide libraries displayed on the surface of filamentous bacteriophage FUSE5 were screened by DNA sequencing. The inserted amino acid sequence. Results After three rounds of screening, some specific phage clones were obtained and 10 clones were randomly selected for DNA sequence analysis. Among them, 8 clones contained 6 peptide sequences and 2 clones contained 15 peptide sequences. Conclusion The specific short peptide with high affinity and strong binding ability to damaged endothelial cells can be screened by phage technique
