目的:观察丹蒌片对动脉粥样硬化(atherosclerosis,AS)模型大鼠血清抵抗素(Resistin)和血管内皮的影响,探讨丹蒌片对AS模型大鼠血管内皮的保护作用及可能机制。方法:32只雄性Wistar大鼠随机分为正常组、模型组、他汀组、丹蒌组,每组8只。正常组大鼠喂食基础饲料12周,余各组均采用高糖高脂饮食+维生素D3腹腔注射方法复制AS模型。模型组、他汀组、丹蒌组,分别给予生理盐水、阿托伐他汀片混悬液、丹蒌片混悬液灌胃干预8周。干预结束后,行ELISA法检测大鼠血清Resistin、内皮素-1(ET-1)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)和一氧化氮(NO)的水平。结果:1与正常组比较,模型组大鼠血清Resistin、ET-1、IL-6和TNF-α水平升高(P<0.05,P<0.01,P<0.05,P<0.05),NO水平下降(P<0.05);2与模型组比较,他汀组大鼠血清Resistin、ET-1、IL-6和TNF-α水平降低,差异有统计学意义(P<0.01,P<0.01,P<0.01,P<0.05),NO水平上升(P<0.01);丹蒌组大鼠血清Resistin、ET-1、IL-6和TNF-a水平降低,差异有统计学意义(P<0.01,P<0.01,P<0.01,P<0.01),NO水平上升(P<0.01);3他汀组与丹蒌组之间大鼠血清resistin、ET-1、IL-6、TNF-α和NO之间水平差异无统计学意义(P>0.05)。结论:1丹蒌片可降低AS模型大鼠血清Resistin、ET-1、IL-6和TNF-α水平,升高NO水平,减轻AS模型大鼠血管内皮的损伤,作用与他汀类药物相似。2丹蒌片可保护血管内皮,其作用机制可能是通过降低血清Resistin、ET-1、IL-6和TNF-α水平,增加NO的分泌,从而保护血管内皮细胞结构的完整和功能的完善。
OBJECTIVE: To observe the effect of dancao tablet on serum resistin (Resistin) and vascular endothelium in atherosclerosis (AS) model rats and to explore the protective effect of dancao tablet on the vascular endothelium in AS model rats and its possible mechanism. Methods: Thirty-two male Wistar rats were randomly divided into normal group, model group, statin group and Dan-Dan group, with 8 rats in each group. The rats in normal group were fed with basic diet for 12 weeks. All the other groups were given high glucose, high fat diet and vitamin D3 intraperitoneal injection for AS model. The model group, the statin group and the Dan-Tan group were given saline, atorvastatin tablets suspension and Dan-Dan tablets suspension for 8 weeks. After intervention, serum levels of Resistin, ET-1, IL-6, TNF-αand NO )s level. The levels of serum Resistin, ET-1, IL-6 and TNF-α in model group were significantly higher than those in normal group (P <0.05, P <0.01, P < (P <0.05) .2 Compared with the model group, the levels of serum Resistin, ET-1, IL-6 and TNF-αin the statin group were significantly lower (P <0.01, P <0.01, P <0.01) (P <0.01, P <0.05), and the level of NO increased (P <0.01). The levels of serum Resistin, ET-1, IL-6 and TNF-a in Danji group were significantly decreased , P <0.01, P <0.01), and the level of NO increased (P <0.01). The difference of serum resistin, ET-1, IL-6, TNF- No statistical significance (P> 0.05). Conclusion: Dan Dan tablet can reduce the level of serum Resistin, ET-1, IL-6 and TNF-α in AS rats, increase the level of NO, and attenuate the injury of vascular endothelial cells in AS rats, which is similar to that of statins. 2 Dan tablets can protect the vascular endothelium, and its mechanism may be through the reduction of serum levels of Resistin, ET-1, IL-6 and TNF-α, increase the secretion of NO, thereby protecting the integrity and function of vascular endothelial cell structure.
