目的:通过比较未经灌流处理和体外灌流处理的慢性重型肝炎患者血浆对C3A细胞CYP4503A表达的影响,为改善C3A细胞的功能活性以提高其治疗效果提供依据。方法:分别用正常人血浆、慢性重型肝炎患者血浆、体外灌流处理的慢性重型肝炎患者血浆培养C3A细胞,RT-PCR法测定上述血浆培养的C3A细胞CYP4503A4 mRNA表达,凝胶成像系统获取CYP4503A4/β-action相对值,进行统计学分析。结果:未经灌流处理和体外灌流处理的慢性重型肝炎患者血浆培养的C3A细胞CYP3A4 mRNA表达均低于正常人血浆培养组(P<0.05),而体外灌流处理的血浆培养的C3A细胞CYP3A4 mRNA表达高于未经灌流处理的血浆培养组(P<0.05)。结论:在进行生物人工肝治疗之前,配合血液灌流技术对患者血浆进行适当的处理,从而改善生物反应器中细胞的功能活性,将可能提高生物人工肝的治疗效果。
OBJECTIVE: To compare the effect of plasma on CYP4503A expression in C3A cells in patients with chronic severe hepatitis treated by perfusion and in vitro perfusion, and to provide evidence for improving the functional activity of C3A cells and improving their therapeutic effect. Methods: C3A cells were cultured in plasma from patients with chronic severe hepatitis and plasma from patients with chronic severe hepatitis, respectively. The expression of CYP4503A4 mRNA in C3A cells was detected by RT-PCR. The CYP4503A4 / β -action relative value, for statistical analysis. Results: The expression of CYP3A4 mRNA in C3A cells was lower in patients with chronic severe hepatitis than those in normal controls (P <0.05). However, the CYP3A4 mRNA expression in C3A cells cultured in vitro Higher than those in the non-perfusion group (P <0.05). CONCLUSIONS: Prior to the bioartificial liver therapy, appropriate treatment of the patient’s plasma with hemoperfusion technique may improve the functional activity of the cells in the bioreactor and may improve the therapeutic effect of the bioartificial liver.
