目的探讨流式细胞术检测CD4+CD25+叉头状转录因子(Foxp3)+天然调节性T细胞的方法及其在系统性红斑狼疮(SLE)患者外周血中改变的意义。方法建立三色流式细胞术检测天然调节性T细胞的方法,并检测了15例SLE患者和15名健康对照者外周血CD4+CD25+Foxp3+调节性T细胞比例以及CD4+细胞中CD25的表达水平。结果SLE患者外周血中天然调节性T细胞为(1.03±0.42)%,显著低于健康对照组的(5.88±1.40)%(P<0.01);CD4+淋巴细胞中CD25+细胞的表达水平[(5.92±3.80)%]也显著低于健康对照组[(32.01±5.70)%,P<0.01]。结论三色流式细胞术适合检测外周血中天然调节性T细胞,SLE患者存在天然调节性T细胞免疫缺陷。
Objective To investigate the method of flow cytometry for the detection of CD4 + CD25 + Foxp3 + natural regulatory T cells and its significance in the peripheral blood of patients with systemic lupus erythematosus (SLE). Methods Three-color flow cytometry was used to detect natural regulatory T cells. The proportion of CD4 + CD25 + Foxp3 + regulatory T cells in peripheral blood and the expression of CD25 in CD4 + cells were measured in 15 SLE patients and 15 healthy controls . Results The percentage of natural regulatory T cells in peripheral blood was (1.03 ± 0.42)% in SLE patients and significantly lower than that in healthy controls (5.88 ± 1.40)% (P <0.01). The levels of CD25 + cells in CD4 + lymphocytes ± 3.80)%] were also significantly lower than those in healthy controls [(32.01 ± 5.70)%, P <0.01]. Conclusion Three-color flow cytometry is suitable for the detection of natural regulatory T cells in peripheral blood. SLE patients have natural regulatory T cell immunodeficiency.
