目的建立以HPLC-ELSD对黄芪提取物中单糖和二糖进行测定的方法。方法采用PrevailTMCarbohydrate ES色谱柱,乙腈-水(75∶25)为流动相,柱温35℃,流速0.8 mL.mim-1;ELSD漂移管温度60℃,载气为空气,气压25 psi。结果黄芪提取物中的单糖和二糖组分能够得到较好地分离,果糖和蔗糖的线性范围分别为0.052~1.042、2.442~48.84 mg,r分别为0.9947、0.9965;平均加样回收率分别为98.6%、102.9%,RSD分别为1.2%、1.0%。结论所用方法快速简捷、结果准确稳定,适合于黄芪提取物中单糖、双糖的含量测定。
OBJECTIVE To establish a HPLC-ELSD method for the determination of monosaccharides and disaccharides in astragalus extract. Methods PrevailTMCarbohydrate ES column was used. The mobile phase was acetonitrile-water (75:25). The column temperature was 35 ℃ and the flow rate was 0.8 mL.mim-1. The ELSD drift tube temperature was 60 ℃. The carrier gas was air and the pressure was 25 psi. Results The monosaccharide and disaccharide components of astragalus extract could be separated well. The linear ranges of fructose and sucrose were 0.052 ~ 1.042,2.442 ~ 48.84 mg, r were 0.9947 and 0.9965, respectively. The average recoveries were 98.6%, 102.9%, RSD 1.2%, 1.0% respectively. Conclusion The method is rapid and simple, accurate and stable, suitable for the determination of monosaccharides and disaccharides in Astragalus extract.
