测定了光敏剂血卟啉单甲醚对人宫颈癌细胞HeLa光敏作用后的傅里叶红外光谱。结果显示:光敏作用后,HeLa细胞磷酸二酯基团的对称伸缩振动峰1085cm~(-1)和不对称伸缩振动峰1246cm~(-1)蓝移,强度下降:蛋白质酰胺Ⅰ带1656cm~(-1)发生蓝移,酰胺Ⅱ带1546cm~(-1)出现红移;CH_2对称伸缩振动峰2858cm~(-1),峰位蓝移2cm~(-1),峰值明显减弱:细胞的蛋白质和核酸谱峰面积比值D1085/D1546降低。提示细胞中的DNA、蛋白质和磷脂结构受到损伤。结果表明:DNA、蛋白质和磷脂是血卟啉单甲醚光敏作用的主要靶分子。
The Fourier transform infrared spectroscopy (FT-IR) of photosensitizer hematoporphyrin monomethyl ether on human cervical cancer cell HeLa was measured. The results showed that the intensity of the symmetrical stretching vibration peak 1085cm -1 and the asymmetric stretching vibration peak 1246cm -1 of HeLa cells decreased with the increase of photosensitizing intensity. The intensity of the protein amide Ⅰ band was 1656cm ~ (-1) -1), the red shift of the amide Ⅱ band at 1546cm -1 was observed. The peak of symmetry stretching vibration of CH_2 was 2858cm -1 and the peak blue shift was 2cm -1. And nucleic acid peak area ratio D1085 / D1546 decreased. Tip cells in the DNA, protein and phospholipid structure is damaged. The results showed that DNA, protein and phospholipid were the main target molecules of hematoporphyrin monomethyl ether.
