用硝酸钙添加剂形成难熔的氟化钙,高温灰化进行样品前处理。以柠檬酸钠—氯化物—冰醋酸所组成的总离子强度调节剂(TISAB)的pH值控制在5.0~5.5之间,用氟离子选择电极测定生物样品中氟含量,间接求出5-FU、FT-207。可测血浆氟线性范围0.005~0.05和0.05~20μg/ml。相关系数为0.9998和0.9991,检测下限0.005μg/ml.血浆5-FU可测范围0.00~6840.39μg/ml。测定血浆中氟及5-FU标准差为0.005、0.08;变异系数为2.3%、4.5%;回收率为97.5%、84.0~111.2%。对大剂量口服FT-207的狗,测定内脏FT-207,获满意结果。本法也适用于其它含氟药物的测定
A refractory calcium fluoride is formed with calcium nitrate additive and the sample is pre-treated at high temperature ashing. The total ionic strength regulator (TISAB) composed of sodium citrate-chloride-glacial acetic acid is controlled at a pH between 5.0 and 5.5. Fluorine ion selective electrode is used to determine the content of fluorine in biological samples. Out of 5-FU, FT-207. Measurable linear range of plasma fluorine 0.005 ~ 0.05 and 0.05 ~ 20μg / ml. The correlation coefficient was 0.9998 and 0.9991, the detection limit was 0.005μg / ml. Plasma 5-FU can measure the range of 0.00 ~ 6840.39μg / ml. The standard deviation of plasma fluoride and 5-FU was 0.005, 0.08; the coefficient of variation was 2.3% and 4.5%; the recoveries were 97.5% and 84.0 ~ 111.2% respectively.
