目的研究丹参脂溶性成分对大鼠视网膜Müller细胞的毒性作用。方法以不同浓度的丹参脂溶性成分及丹参酮ⅡA对照品分别干预体外纯化培养的大鼠视网膜Müller细胞48 h,采用CCK-8法检测其细胞增殖抑制率。结果测得丹参脂溶性成分及丹参酮ⅡA对照品对大鼠视网膜Müller细胞增殖均有一定抑制作用,且具有剂量依赖性。CCK-8法测得丹参脂溶性成分及丹参酮ⅡA对照品的IC50分别为2.181、6.176μg·mL-1,给药20μg·mL-1时,测得细胞增殖抑制率分别为75.8%、58.7%。结论丹参脂溶性成分及丹参酮ⅡA对照品对大鼠视网膜Müller细胞均有一定毒性,所测IC50值为进一步筛选研究丹参脂溶性成分治疗糖尿病性视网膜病变作用机制的有效浓度提供了一定的实验依据。
Objective To study the toxic effects of lipophilic components of Radix Salviae Miltiorrhizae on retinal Müller cells in rats. Methods Different concentrations of Salvia Miltiorrhiza fat-soluble components and tanshinone Ⅱ A reference substance were cultured in vitro purified rat retinal Müller cells for 48 h, using CCK-8 assay inhibition rate of cell proliferation. Results The lipid-soluble constituents of Salvia miltiorrhiza and the reference substance of tanshinone Ⅱ A were found to inhibit the proliferation of retinal Müller cells in a dose-dependent manner. The IC50 of lipid-soluble constituents of Salvia miltiorrhiza and the reference substance of tanshinoneⅡA measured by CCK-8 method were 2.181,6.176μg · mL-1, respectively, and the inhibitory rates of cell proliferation were 75.8% and 58.7% at 20μg · mL- . Conclusion The lipid-soluble constituents of Salvia miltiorrhiza and the reference substance of tanshinone ⅡA are toxic to retinal Müller cells in rats, and the IC50 values provide some experimental evidence for further screening of the effective concentration of lipid-soluble components of Salvia miltiorrhiza in the treatment of diabetic retinopathy.
