目的:建立LC-APCI-MS法测定血浆中西咪替丁的血药浓度。方法:采用Zorbax SB-C18(2.1mm×100mm,3.5μm)色谱柱;流动相为甲醇-0.1%甲酸(60∶40)。大气压化学电离源(APCI),选择正离子模式检测,采用选择离子监测(SIM)方式进行定量分析,检测离子为m/z253(西咪替丁),m/z180(内标非那西丁)。结果:血浆中西咪替丁在10~2 000μg.L-1范围内线性关系良好,最低检测限为10μg.L-1,西咪替丁的平均萃取回收率为74.2%~75.1%,日内及日间RSD都低于13%。结论:该方法操作简便、灵敏度高,可用于西咪替丁的临床血药浓度监测。
Objective: To establish a LC-APCI-MS method for the determination of plasma cimetidine plasma concentration. Methods: Zorbax SB-C18 (2.1mm × 100mm, 3.5μm) was used as the mobile phase. The mobile phase was methanol-0.1% formic acid (60:40). Atmospheric pressure chemical ionization source (APCI), positive ion mode detection, selective ion monitoring (SIM) quantitative analysis, the detection of ions m / z253 (cimetidine), m / z180 . Results: The linearity of cimetidine in plasma was good in the range of 10-2000μg.L-1 with the lowest detection limit of 10μg.L-1, the average extraction recovery of cimetidine was 74.2% -75.1% Daytime RSDs are below 13%. Conclusion: The method is simple, sensitive and can be used to monitor clinical plasma concentration of cimetidine.
