目的:了解采用机械搅拌法提取的粘性放线菌Ⅰ型、Ⅱ型两种菌毛的化学组成。方法:首先制备出抗Ⅰ型菌毛抗血清和抗Ⅱ型菌毛抗血清,鉴定两种菌毛纯化物纯度后,采用考马斯亮蓝法和蒽酮法分别测定两种菌毛中蛋白质和糖的含量,并进行氨基酸组成分析。结果:两种菌毛的主要成分是蛋白质,糖含量较低。两种菌毛均是天门冬氨酸、谷氨酸、丙氨酸、赖氨酸和甘氨酸含量较多。两种菌毛中,非极性氨基酸百分含量最高,碱性氨基酸百分含量最低。结论:本实验结果有助于了解两种菌毛的化学组成与其生物性能的关系。
OBJECTIVE: To understand the chemical composition of two types of pili actinomyces viscosus Ⅰ and type Ⅱ extracted by mechanical agitation. Methods: Firstly, the anti-type pilus antiserum and anti-type pilus anti-serum were prepared. After the purity of the two kinds of pilus purified was identified, the protein and sugar in the two kinds of pilus were determined by Coomassie brilliant blue method and anthrone method respectively Content, and amino acid composition analysis. Results: The main components of the two pili were protein with low sugar content. Both pili hair are aspartic acid, glutamic acid, alanine, lysine and glycine more content. The two pili hair, the highest percentage of non-polar amino acids, the lowest percentage of basic amino acids. Conclusion: The results of this experiment help to understand the relationship between the chemical composition of two pili and their biological properties.
