目的:对THO1,TPOX和D6S1043,D12S391基因座在北方汉族群体中的法医学应用价值进行比较。方法:用chelex100法提取445名北方汉族无关个体血痕的DNA,分别采用Identifiler和Sinofiler系统复合扩增,用ABI3130XL基因分析仪进行毛细管电泳,GeneMapper IDv3.2软件进行分型,计算四个基因座的多态信息。结果:四个基因座基因型分布符合Hardy-Weinberg平衡。统计结果计算显示D6S1043、D12S391、TH01和TPOX基因座的杂合度(H)分别为0.87、0.845、0.67和0.662:个人识别能力(DP)分别为0.968 6、0.951 0、0.856 2、0.805 6:多态信息含量(PIC)分别为0.861、0.816、0.633、0.576,非父排除率(PE)分别为0.682 4、0.635 0、0.382 4、0.373 3。结论:D6S1043和D12S391在北方汉族人群中具有良好的多态性,更适合应用于北方汉族人群的个体识别与亲权鉴定。
Objective: To compare the forensic value of THO1, TPOX, D6S1043 and D12S391 loci in northern Hans population. Methods: The DNA of 445 unrelated Han individuals was extracted by chelex100. The DNA was amplified by using Identifiler and Sinofiler system respectively. The DNA was analyzed by capillary electrophoresis with ABI3130XL gene analyzer and GeneMapper IDv3.2 software. Polymorphic information. Results: The four loci genotype distribution in line with the Hardy-Weinberg equilibrium. The statistical results showed that the heterozygosities (H) of D6S1043, D12S391, TH01 and TPOX loci were 0.87,0.845,0.67 and 0.662, respectively. The personal identification ability (DP) was 0.968 6,0.951 0,0.856 2,0.805 6: (PIC) were 0.861,0.816,0.633,0.576 respectively, and the non-parent exclusion rate (PE) was 0.682 4,0.635 0,0.382 4,0.373 3 respectively. Conclusion: D6S1043 and D12S391 have good polymorphism in Northern Han population, and are more suitable for individual identification and paternity test in Northern Han population.
