临界点干燥是制作扫描电镜生物标本的重要环节。我们对日立HCP—2型临界点干燥器的临界现象进行了实验。实验表明:每次干燥样品的临界现象与其CO_2的充液量及纯度有关。一、实验步骤 (室温在20℃±2): 1.干燥室预冷到5—10℃,迅速放入标本。 2.缓慢充进CO_2,待2~3分钟后,打开微放气阀门(气柱高为2cm),放气3~5分钟。
Drying of the critical point is an important part of the production of scanning electron microscopy biological specimens. We conducted an experiment on the critical phenomena of Hitachi HCP-2 critical point dryer. The experiment shows that the critical phenomenon of each dry sample is related to the filling amount and purity of CO 2. First, the experimental steps (room temperature at 20 ℃ ± 2): 1. The precooling room to 5-10 ℃, quickly into the specimen. 2. Slowly filled with CO_2, to be 2 to 3 minutes later, open the micro-deflation valve (column height 2cm), deflated 3 to 5 minutes.
